Stake a claim on their lands, but that will be a slow and gradual process. And assassinations work too.
But assassinations won't do much by themselves, as their heir will be another one of the dynasty. Also don't think you can stake a claim on a kingdom title.
But what did work was to start a faction for elective succession, and when they were in a civil war, I stroke. I won, then nominated myself as successor, saw I was in 1st place, and THEN assassinated the ruler :)

Great job! I just meant to slowly engulf the Westerlands in many wars for small pieces of land. And assassinate just to throw them off so the rulers are constantly weaker.
 
Is it possible to, say, take The Westerlands from the Lannisters without mixing my blood with theirs?

EDIT: My bad I thought this was about the iron throne but the jist of this applies to the other 7 kingdoms. You'll need to wait for a mega war or play before the Aegon's Landing.

Theres a number of ways that you can do it but most of them are lore breaking and almost all of them gamey.

1. The easiest thing, (or fastest rather), I can think of is "Overthrow the Monarchy" faction, raise all your levies as one of the powerful 7 kingdoms. Plant all your levies on King's Landing. Demand the government conversion, they'll almost always go to war if you don't have a huge amount of support. Get lucky and capture the king/heir/whoever and sue for peace. Stay elected for ten years and with high diplomacy remake it into a monarchy lol

4. Emancipation CB with liberator maybe?

6. Dragon CB (So OP)

7. Use another empire, (e_north, e_westerlands etc etc) and conquest the e_iron_throne then make it your primary title and elect lord paramounts for your former territory. I'm not sure if this works exactly but it probably does.
