make sure your destination as less than 5000 inhabitants, click you colonization button (the one with the boat) and then click on the province (it should be dark green) and then click send colonist and voila!
Colonists are missionaries by default. If the destination has another religion they will always convert them. Evidently u can't send a settler before the criteria are met (like less than 5000 inhabitants)
