So, there are 3 shifts, an no one likes to work the night shift.

So, lets say for the purposes here, there are only 2 shifts.

1) If I employ shop staff to work the morning shift, and no one in the afternoon, does that mean no one can access the shop services in the afternoon?

2) If I have 100 employment, and everyone is assigned to work the morning shift, and everyone is "free" in the afternoons, does this mean no one gets to use the services at all? As all people are either busy working (morning shift) or everywhere is closed (afternoon shift),

Or do these things work in a different way?
 
No i think you are correct. Try to staff service building for the whole day, so everybody has a chance to attend it. There is also a limit on how many people can use a service building at the same time. I guess the limit on the building-tooltip is per hour, but i'm not sure (might also be per shift).
So having only one grocer in a dome full with apartments might not be enough.
