This is indeed also something I noticed after the lates patch/DLC. Scheduled vehicles (buses, trams, trolleys etc) just randomly stop.
 
It's a bug that has actually never been resolved. Besides countless save games that had been provided. It appears when you add or delete roads. The developers claim they are not able reproduce it.....uhm yes...
I gave up on it, but the bug destroys a game.

That was a different issue. It stopped the entire traffic, but now it only stops buses, trams etc.
 
Perhaps... Still, the result is pretty much the same. This bug happened in my game with the trams and trolleys, which makes me think that similar to the issue with the cars getting stuck on account of roads having been changed, this is a direct result of a tram/trolley line being edited while there is at least one active tram/trolley using that particular line. One could argue that in real life an active line would never be edited on the fly but I doubt if returning all vehicles to the depot beforehand would change anything.
 
This is happened with me, making a bus queue and causing a traffic, so, the depots were empty and the bus stations were crowded, but they would not budge, so I had to put a sidewalk parallel to intercede and remove the vehicles.
 
I had something similar yesterday. I left the game for months but i'm back now, thanks to the two new vehicle sets.
I was able to solve this problem with simply deleting the faulty road piece and replacing it. Now it's working fine. Annoying bug, but if you have some patient you can solve it most of the time.
I often modify the roads near my transportation hubs, so it's kind of expectable (i know, we should not expect bugs like these, but still)
