Last time I tried it 4 years ago, it didn't work (even if assumed to be supported).
So, is it possible to change defines through game rule, directly or through event fired with said rule, today?

No and it never will be. Those that can be changed dynamically have already been moved to script_values. Defines are values used directly by the game engine that for whatever reason need to be constant and static.
