Hi

I recently bought Cities Skylines from the Microsoft Store and I can't work out how to get the Content Manager - there's no option for it in the main menu. Do I need to activate it somehow or do I need to buy it separately?

Here's a screenshot of what I see.

The Microsoft Store edition doesn't come with the Content Manager or the ability to add custom content. Practically it's the same version of the game as consoles get. The PC version that can use custom content is the one you get of Steam or Origin. I'd recommend getting it through Steam, as Origin doesn't have access to the workshop.
