This bug can easily be seen if you load up a game as Russia in the 1617 scenario (the age of mercantilism.) There are two Russian provinces with "Ugric" culture, both with natives and both with less than 700 people. Sending a colonist to either should change the culture to Russian (and the religion to Orthodox,) however one will not change - the one that gets turned in to a city because of the larger population and the natives.
I believe this is a bug in the order the game calculates whether or not to convert a provinces culture - it should be done before the natives are added into the resulting colonial city, and not the other way around. This should help out the frontier nations a little bit if fixed (and help eliminate the confusion regarding cultural transformations that exists now.)
