So, I'm in the middle of my first game, but I've gotten paranoid about not having re-supply points on my routes, to the point where I've signed pacts with several nations, and I've conquered the Canaries. I think of this as getting your army stuck in enemy territory after a peace treaty, without having military access back to your own country, to give an EU example. So my questions are:

How exactly does not having an available port to re-supply in between Home port and an Indian port affect you? Could I use this as a tactic to choke out an enemy company?
 
It slows your fleet to a real crawl until it can resupply again. I usually have Pacts signed with everyone I'm not at war with. I also never take ports like Canaries which are basically useless except for resupply.

You can always give Canaries to a friendly AI company with whom you have a pact. This way they pay the upkeep and you still retain access.
 
I like having all the close ports.

I minimize their upkeep by destroying most of theier buildings. The benefit is that if you dont pact with anyone, everyone else has to sail really slowly to thier home port giving you a big advantage.

I am not sure how this affect combat...but i would atleast think your fleets that are out of supply would fight with atleast a moral loss or something. Haven't checked but this could be a real sweet way to reduce anyone at war with you by only fighting battles in this area so they are out of supply.
 
AI is more likely to attack ports in the area where their ships get crawling so it's hard to keep such a state.

And I don't think fleets are any different on the tactical level (or in auto-resolve) being out of supplies. They could though so maybe you should post this to the ideas sticky thread?
