The only way I can think of is shenanigans around country rank (such as going into the next one to get a new family, then dropping down and losing an old one).
 
Another Q before the new game: is there anyway to get rid of/replace/kill off a major family? Or will they always adopt new characters to keep up the line?
Yes - imprison the family head by putting him/her to trail, then proscribe. Be warned though: Proscribtion both requires substantial Tyranny (40) before it is available - and then further piles on that score (+30 Tyranny). One effect of the proscribtion is that the great family is no longer, all former mebers become minor characters and the rise-from-obscurity event will trigger next month, which lets you pick a new great family.

Just wondering for immersion purposes I suppose. For example, if as Rome I take a liking to a particular minor character would have liked to see them rise to the top since I can’t actively adopt as Rome. No biggie if this isn’t really a possibility though.
It's not uncommon for family heads to become consul (or vice versa), but yes there's no way to sponsor a minor character into major family status.
 
I belive that while in Republics the human player can't adopt into the own family, the AI family heads still can do so (and will, though only in the case that they have shrinked so much that not enough adult characters are left to fill their amount of requested jobs); usually they pick minor characters holding offices.
