Please explain your issue is in as much detail as possible.
I created a map and successfully uploaded to Steam. I deleted local copy and subscribed to the one on Steam. That part worked.

Now I made some changes to the map and wish to update on Steam, but saving the map creates a new copy. Clicking "Update" button does nothing, my changes are not uploaded.

Why would you delete the local version of your map? To be able to successfully update on Steam you will need your local version. No way around it.
 
When you click update on the map you're subscribed to, you need to open then content folder and copy the updated/local version of the map into the content folder. Otherwise, it's just "updating" with the existing map, not the new version.

There's a step-by-step guide here. It's a little old so the interfaces have changed a little, but the principles are the same as they originally were. Hope that helps!
