Please explain your issue is in as much detail as possible.
Shortly after my character (King of England) got married, all members of my family have disappeared from both the "Family" tab and the Family Tree. I'm only one year into the game with my first character. Can they be restored, or do I have to start over?

I've opened the game with just the DLCs you've mentioned. The King of England at the start of the game is AEthelstan with the of Wessex dynasty. However, your picture shows that your character is an Eanfrith with the Northampton dynasty. There are only two Eanfriths that exist in the 936 start date, the Captain of the White Company and the Bishop of Southwark, both of whom are lowborn with no claims. Since neither has a claim nor are playable, your character can't be either of them. Then, you must have used the Ruler Designer.
I created a character with the Intricate Webweaver education and the Wounded (common Ruler Designer trick), Scholar, Genius, and Stutter Traits and he just so happened to be 48 years old. One year later in-game is 49 years old.

Now, when you use the Ruler Designer, you completely override the existing character with those titles. You don't keep their siblings, parents, grandparents, or children (although you can add children at the cost of starting two or three years older per child).

Now, is it possible you just didn't notice your character had no family for 1 year? If so, not a bug. Alternatively, if you created a character with the Ruler Designer and had family without choosing children in the Ruler Designer, that would a bug.
 
Well, let's just say that I'm 90% sure that I had parents and grandparents at the start of the game --- or at least I was until your thorough explanation of what likely happened. This is only my first real game, so I was definitely playing slowly and looking at everything. I was unaware that using the Ruler Designer left you with no ancestors, which I guess isn't usually a big deal.

I'm pretty sure that isn't correct. You don't override the existing ruler, you just displace him. He and his family member that were in his court continue to exist, just as unlanded courtiers in your court.
 
I think you're both correct. AEthelstan did continue to exist (although I didn't check his family), and in fact there was a powerful faction to put him back on the throne that I had to deal with! But it does appear that using the Ruler Designer doesn't also give you any ancestors.
