I tried to do one with an 'Additional Inventory : 8 Drones' requirement. I spent a while trying to figure out how to get drones onto the rocket - the inventory showed 0/8. I have 10 drones in my colony. I tried assigning them to the rocket and packing them into prefabs. There was some fuel that needed to be unloaded, once it unloaded the rocket took off (still with 0/8). It just came back and dropped off a bunch of drones - the rocket started with 8/8 drones and now has 2/8 drones. The 6 drones that came out are assigned to the rocket but not doing anything. The rocket is currently waiting for 29 polymers to unload (presumably that was from the mission)? But the rocket info panel still says "This Rocket is going to analyze a Planetary Anomoly'. So I'm pretty confused on how it is supposed to work because this doesn't feel right.

Summary:
  1. Rocket took off with 0/8 drones
  2. Rocket came back with 8/8 drones, dropped two off
  3. Those 6 drones are doing nothing and the other 2 aren't coming off the rocket
  4. The rocket still indicates it is on a mission

Did you try the unload the 29 Polymers and see if the Rocket will return to normal mode. Seems to me that it's stuck right now, trying to unload something and failing to do so. Please, if you have a saved game, send us an in-game bug report with a savegame (Ctrl+F1).

About your question, the launching procedure for expeditions to planetary anomalies and rival colonies is as it follows:
1. The Rocket loads fuel and unloads excess cargo.
2. The Rocket loads the required payload (drones/rovers/colonists).
3. The Rocket launches.
 
I Had a similar problem, but I believed it was through mods. ... maybe I can reproduce it

Do the drones need to be packed as prefabs to load or should they load themselves?
 
They don't need to be packed, and should just load themselves.
I'm not sure how close to the rocket they have to be. They just do it all themselves once the rocket is ready.
