If I purchase it on Steam and download, can I later run it offline and with no Steam running via a file in the game's folder? I'm interested in the linux version.

I am sure i've played it before without internet connection. As long as you download it and activate it online you can then play it offline from then on I think. (just checked it and it does work offline)
you just can't take advantage of the cloud storage for saves.
 
You can only start the game when steam is running. But Steam can be set into offline mode, so no connection to the internet is necessary. Also the Linux-version doesn't start without steam running.
 
Nearly all games sold in Steam are not DRM free per definition, since SteamWorks is a DRM system. Some games start without Steam having to run in the background. However, this is only a small minority.
For CiM2, as mentioned before, Steam definitely has to run.
However, as pointed out, Steam has an offline mode. Therefore, you only need a internet connection once to activate and download the game. Afterwards you can set Steam to offline mode and start CiM2.

Not all games on Steam have SteamWorks as thier DRM.
Some games have third-party DRM, some don't have DRM at all. CiM1 for example can be started from it's exe file without any problems, even when Steam is shut down. CiM2 must have Steam running, and must be started from Steam Client. Offline mode will allow you to play without an internet connection.
