Should slave estates increase slave ratio in the settlement it is built?

I think you build it because you want slaves there?

Or am i missing something?

I would actually say no. Slaves can potentially generate a lot of unrest, so while having a really high slave proportion might seem like a good idea, it's going to tank province loyalty and generally be quite annoying to manage. Freemen are okay pops, and in most civilized governments they're fairly easy to keep happy which means a balance of slaves and freemen will generally be fairly managable.

The desired ratio is a up-bottom decision to modifiy POPs composition in your territory. But even then, devs have had the thought to describe that slaves would not want to move to slave states as they want to promote and break free.

I am against desired ratio to decide POPs in a territory but not having increased slave ratio in slaves states is fine.
