Thank you for the report. Does this also happen if you play without any mods?
To disable mods, please try adding "--disableModding" as a launch option (https://pdxint.at/LaunchOptionsCS). This removes access to PDX Mods entirely, and if the game and a new save works with this enabled then the issue lies in the mods you have installed, and if you're unable to disable your mods in-game you can always do so on the web version.

I can confirm that without mods it still doesnt work. specialized industry still doesnt send trucks out
 
I have found they do send out trucks but it takes a very, very long time for it to happen. They are not able to sell enough material to make a profit and that keeps them at lvl 1 and always losing money.

guess i didnt wait long egnough, altough i dont think this is the way they are supposed to work. and still is a bug
 
I was researching this issue today, and come across this thread that pretty extensively dives into this problem, and other related symptoms that were theorized to be related. (Thanks joewagner501, who is on this thread, for the awesome troubleshooting). It seems like the specializations are very slow to harvest, and when they do, they don't export their goods so they never make a profit and thus stay level 1.
I've tried fresh saves, no mods, different sizes of harvestable area, different specializations, forcing the buildings to level through dev-mod, placing the farms on different land values, making sure there no two specializations were placed in the city, and waiting for several IRL days for the buildings to export and trying different tax levels for each specialization type to see if it helps but nothing has worked. I'm happy to provide a save for further troubleshooting if that helps!

I see same issues. I have all specialized industries set to -10% in taxes and they are stuck at an intermediate level or at 1. For agriculture, I have rezoned as cotton, vegetables, or grain on most fertile area and they never increase. Service fees on electricity and water at 0%. They all come in at ~140% efficiency but never level up. Oil is the same as well.

If they do, it's extremely rare. Every single time I've clicked on a building to check, it says 0 vehicles in use. I even placed a cargo port right next to my farms but I haven't seen them make a single delivery for export. Of course it's still possible they are sending out trucks since I'm not checking on them 100% of the time, but like you said, if they do it's not nearly often enough to make any money.
 
Through extensive testing I think the issue is two-fold.

The first is the specialized industry doesn't export very often even when a freight shipping (rail/port) is placed in the city. I also never see the local zoned industrial buying directly from them. I see zoned commercial buying directly from zoned industrial but that is it. Zoned industry doesn't buy from specialized industry. I think both should be looked at. It may be the local buying not taking place that could be the major issue. I think this because if there is no local commercial buying from local industry, then that industry business doesn't make a profit even if it's exporting product occasionally.
 
Not sure if exactly the same issue but noticed my specialized industry mostly never levels up but does fall into the trap of endless going from vacant to occupied for the same companies.

This being a test that to my eyes is the best situation I can put them in to succeed:

Vegetable farms where there's a deficit and need for imports, plenty of cargo export options, zero service fee for electricity and water, -10% industrial taxation.

Land value across the three in the clip goes from ¢85 at the nearest farm, ¢30 for the middle, and down to ¢10 for the one at the back - showing no difference.

What am I missing here...
 
I sat and watched an industry that required vegetables and livestock to produce food. It was a level 3 zoned industry with a profitability of "getting by (50%). I wanted to see what happened when the "Requires" stock dropped to see when it would be replenished and how it would be replenished.

What I saw was that the business flipped to a new one that required livestock to produce convenience food and because it was a "new" business, it started off with a full amount of livestock. I think this is the underlying problem with the unprofitability of the industry both specialized and zoned. If the businesses keep being turned over, then there is never a need for the "raw materials" of production meaning the specialized industry will never make a profit because there is no industry to sell to since they keep going out of business with a new one taking it's place and starting with a full amount of raw material.
